Assay principle: sandwich
GAD65 molecules are coated onto the ELISA plate wells. GAD65-biotin complexes constitute the liquid phase. GAD-Ab from the samples bind to the GAD65immobilized onto the well and interact with GAD65-biotin via a GAD-Ab bridge. Addition of streptavidin peroxidase detects the bound GAD65-biotin. The unbound streptavidin peroxidase in excess is washed off before the addition of the peroxidase substrate TMB.
Assay procedure: 1h at RT / shaking + 1h at RT / shaking + 20 min at RT / shaking + 20 min at RT in the dark.
